Losing an RNA editor switched on liver scarring through a mitochondrial pathway — in mice and cells

An epitranscriptomic chain — marks on RNA, not changes to DNA — connects a missing editor enzyme to scar tissue, and mitochondria sit in the middle of it: when the chain fired, mitochondrial function faltered and the cleanup of damaged mitochondria changed as the scar cells switched on.
What is the FTO-m6A-IGF2BP1-TRIB3 axis?
FTO removes m6A marks from RNA. In the fibrosis model, FTO fell, m6A levels rose, and the m6A reader IGF2BP1 increased expression of TRIB3, a stress-response protein. The authors describe this FTO-m6A-IGF2BP1-TRIB3 axis as the mechanistic spine of the paper, demonstrated by knocking down or overexpressing FTO and IGF2BP1 in human LX-2 hepatic stellate cells and tracking protein, RNA-methylation and mitochondrial readouts.
Where do mitochondria fit?
FTO depletion was accompanied by mitochondrial dysfunction and mitophagy-related alterations alongside stellate-cell activation — the paper measured mitochondrial function directly as part of its assay panel (histology, immunostaining, western blotting, MeRIP-qPCR and mitochondrial functional assays). The study positions mitochondrial homeostasis as part of how this RNA-methylation pathway drives activation, though it does not claim mitochondria are the sole mediator.
Is this a treatment lead?
The authors call the axis a potential target for epigenetic intervention in liver fibrosis. That is a target nomination from a mouse surgical model and a human cell line — the standard first step, several stages from a drug. No intervention was tested in animals for benefit, and nothing here changes care for people with liver disease.
Frequently asked questions
What is FTO and what does it do?
FTO (fat mass and obesity-associated protein) is an enzyme that removes m6A methyl marks from RNA — an epitranscriptomic editor that helps control how much protein is made from marked transcripts.
What did this study find in liver fibrosis?
FTO was downregulated in fibrotic liver models. Its loss raised m6A methylation, which increased TRIB3 expression through the reader protein IGF2BP1, accompanied by mitochondrial dysfunction, altered mitophagy and activation of scar-producing hepatic stellate cells.
Is there a drug based on this?
No. The authors nominate the pathway as a potential target for future epigenetic intervention. The work is in mice and a human cell line, with no intervention tested.
Where the evidence stands
- Established: Hepatic stellate cell activation drives liver fibrosis; m6A is the most common internal mRNA modification and FTO is one of its demethylases; mitochondrial dysfunction accompanies stellate-cell activation.
- Preliminary: In bile-duct-ligated mice and human LX-2 cells (Scientific Reports, Oct 25, 2026), FTO downregulation raised m6A, increased TRIB3 via IGF2BP1, and was accompanied by mitochondrial dysfunction, mitophagy alterations and stellate-cell activation markers.
- Absent: Any tested intervention, human liver-tissue confirmation of the full axis, and clinical relevance data.
